چکیده

The Na,K-ATPase consists of an and -subunit. Moloney sarcoma virus-transformed MDCK cells (MSV-MDCK) express low levels of Na,K-ATPase 1-subunit. Ectopic expression of Na,K-ATPase 1-subunit in these cells increased the protein levels of the 1-subunit of Na,K-ATPase. This increase was not due to altered transcription of the 1-subunit gene or half-life of the 1-subunit protein because both 1-subunit mRNA levels and half-life of the 1-subunit protein were comparable in MSV-MDCK and 1-subunit expressing MSV-MDCK cells. However, short pulse labeling revealed that the initial translation rate of the 1-subunit in 1-subunit expressing MSV-MDCK cells was sixto sevenfold higher compared with MSV-MDCK cells. The increased translation was specific to 1-subunit because translation rates of occludin and -catenin, membrane and cytosolic proteins, respectively, were not altered. In vitro cotranslation/translocation experiments using rabbit reticulocyte lysate and rough microsomes revealed that the 1-subunit mRNA is more efficiently translated in the presence of 1-subunit. Furthermore, sucrose density gradient analysis revealed significantly more 1-subunit transcript associated with the polysomal fraction in 1-subunit expressing MSV-MDCK cells compared with MSV-MDCK cells, indicating that in mammalian cells the Na,K-ATPase 1-subunit is involved in facilitating the translation of the 1-subunit mRNA in the endoplasmic reticulum.

Na+-K+-ATPase expression in alveolar epithelial cells: upregulation of active ion transport by KGF.

We evaluated the effects of keratinocyte growth factor (KGF) on alveolar epithelial cell (AEC) active ion transport and on rat epithelial Na channel (rENaC) subunit and Na+-K+-adenosinetriphosphatase (ATPase) subunit isoform expression using monolayers of AEC grown in primary culture. Rat alveolar type II cells were plated on polycarbonate filters in serum-free medium, and KGF (10 ng/ml) was ad...

Regulation of Na-K-ATPase gene expression by hyperoxia in MDCK cells.

Na-K-ATPase plays a central role in a variety of physiological processes, including ion transport and regulation of cell volume. Our previous data showed that hyperoxia increased the expression of Na-K-ATPase α1 and β1 mRNA in lung type II cells. We similarly show that hyperoxia (≥95% O2 for 24-48 h) increased steady-state mRNA levels in both Na-K-ATPase subunits in Madin-Darby canine kidney (M...

Na,K-ATPase beta-subunit is required for epithelial polarization, suppression of invasion, and cell motility.

The cell adhesion molecule E-cadherin has been implicated in maintaining the polarized phenotype of epithelial cells and suppression of invasiveness and motility of carcinoma cells. Na,K-ATPase, consisting of an alpha- and beta-subunit, maintains the sodium gradient across the plasma membrane. A functional relationship between E-cadherin and Na,K-ATPase has not previously been described. We pre...

KGF prevents hyperoxia-induced reduction of active ion transport in alveolar epithelial cells.

We evaluated the effects of acute hyperoxic exposure on alveolar epithelial cell (AEC) active ion transport and on expression of Na+ pump (Na+-K+-ATPase) and rat epithelial Na+ channel subunits. Rat AEC were cultivated in minimal defined serum-free medium (MDSF) on polycarbonate filters. Beginning on day 5, confluent monolayers were exposed to either 95% air-5% CO2(normoxia) or 95% O2-5% CO2 (h...

The O-glycosylated ectodomain of FXYD5 impairs adhesion by disrupting cell-cell trans-dimerization of Na,K-ATPase β1 subunits.

FXYD5 (also known as dysadherin), a regulatory subunit of the Na,K-ATPase, impairs intercellular adhesion by a poorly understood mechanism. Here, we determined whether FXYD5 disrupts the trans-dimerization of Na,K-ATPase molecules located in neighboring cells. Mutagenesis of the Na,K-ATPase β1 subunit identified four conserved residues, including Y199, that are crucial for the intercellular Na,...
